Does Pregnenolone Increase Estrogen? And How to Raise It Naturally

Pregnenolone is the first steroid your body makes from cholesterol. Progesterone, DHEA, cortisol, testosterone and estrogen all come after it.
That is why people worry that taking it will raise estrogen. The short answer is that nobody has shown that it does, and nobody has properly ruled it out.
Does pregnenolone increase estrogen?
On paper it can. Pregnenolone can become progesterone or DHEA, DHEA can become testosterone, and the enzyme aromatase turns testosterone into estrogen.
But each step is controlled by enzymes in specific tissues. Having more of the first ingredient does not force the last step.
I searched PubMed for trials that gave people pregnenolone and reported what happened to estradiol. I did not find one with that result in its abstract. What the trials do report is this:
- Schizophrenia trial. Patients took pregnenolone, raised in steps to 500 mg a day, for 8 weeks. It was well tolerated. Serum pregnenolone and its calming metabolite allopregnanolone rose, and negative symptoms improved more than on placebo. (Marx 2009)
- Bipolar depression trial. 80 adults took pregnenolone up to 500 mg a day or placebo for 12 weeks. Neurosteroid levels changed a lot in the pregnenolone group. Remission was 61% against 37% on one of the two depression scales. It was well tolerated. (Brown 2014)
So at doses far above what supplements usually contain, the measured rise was in pregnenolone itself and in brain-active steroids. Estrogen was not what the researchers reported.
The honest part
Not reported is not the same as not happening. These were small psychiatric trials, not hormone studies. If you have high aromatase, from body fat, alcohol or inflammation, more of any upstream steroid could in theory end up as estrogen.
The steroid with an actual record of raising estrogen is DHEA, the next one down the line. In a year-long trial of 50 mg of DHEA a day, testosterone and estradiol both rose slightly. That study is in DHEA: 280 people took 50 mg a day for a year.
Pregnenolone is a hormone. Use it with a doctor and with blood tests, not on a guess.

Ray Peat grouped pregnenolone with progesterone and DHEA as the three youth-associated hormones. He described it as the main brain-protective steroid, with a strong anti-stress action. How to increase pregnenolone naturally
Your body makes pregnenolone from cholesterol inside the mitochondria. According to Peat, that conversion depends on thyroid hormone and vitamin A. So the natural route is to supply the raw material and the conditions:
- Enough cholesterol. Eggs, dairy and shellfish. Very low cholesterol leaves less to convert.
- Thyroid function. Enough carbohydrate, enough calories, and no fasting. Low-carb diets lowered T3 within a week in one study.
- Vitamin A. Liver about once a week, eggs and dairy.
- Lower stress. Under stress the same raw material is pulled toward cortisol.
- Light and sleep. Daily sunlight and 8 to 9 hours of sleep.
